Abstract

Bacillus subtilis AAF2 is an endophytic bacterium that has high anti-phytopathogenic activity against Fusarium oxysporum and Sclerotium rolfsii. Anti-phytopathogenic activity can be increased by choosing the right carbon source. This study aims to find the best carbon source in Bacillus subtilis AAF2 fermentation towards anti-phytopathogenic activity. The study was conducted in vitro and the carbon sources used were glucose, fructose, sucrose and lactose. The parameters were the cells number of Bacillus subtilis AAF2, fluctuations of medium pH and the power inhibition against phytopathogens. The results showed the cells log of Bacillus subtilis AAF2 between 6,11 – 14,74 (CFU ml-1); pH fluctuates between 0.80 - 1.10 from the initial pH of the medium (7.0); and the power inhibition against Fusarium oxysporum between 0.00 – 68.60 %, while for Sclerotium rolfsii between 0.00 – 69.40 %. The best carbon source in this study was glucose at 48 hours of fermentation time with cell log of 14,58 (CFU ml-1); pH 6.7; and the power inhibition against Fusarium oxysporum was 68.60 %, and against Sclerotium rolfsii 69.40 %.
